CDK4 and 6 inhibitors and endocrine therapy have significantly improved outcomes in ER+/HER2- advanced breast cancer, and recently, the first CDK4 and 6 inhibitor was approved in combination with endocrine therapy in the adjuvant setting for patients with HR+/HER2-, node-positive, early breast cancer at high risk of recurrence and a Ki-67 score of ≥20%. However, pretreated ER+/HER2- breast cancer remains a challenging area with limited treatment options.
