Detecting molecular alterations driving the development of specific cancers and then targeting these alterations with matched therapies or combinatorial approaches is now the standard of care for many cancers. HER2 alterations have been identified as key therapeutic targets in breast, gastrointestinal, lung, and other cancers, and targeting these alterations with antibody–drug conjugates (ADCs), antibodies, more specific tyrosine kinase inhibitors, and bispecific antibodies are improving outcomes. In addition to HER2, HER3 represents an emerging target in several cancer types for which novel targeted therapies are being developed, and targeting TROP2 is showing promise as well. Given these treatment advances, biomarker testing to identify patients who may benefit the most from HER2-, HER3-, and TROP2-targeted therapies has never been more important.
