The recent transformation of the bladder cancer therapeutic landscape includes the PD-1/PD-L1–targeting immune checkpoint inhibitors for advanced/metastatic bladder cancer, erdafitinib for FGFR mutation–positive bladder tumors, and the antibody–drug conjugates enfortumab vedotin and sacituzumab govitecan in the post–immune checkpoint inhibitor setting. Utilizing these agents in localized disease settings has led to the emergence of novel bladder-sparing and perioperative approaches, including the first regulatory approval of adjuvant immunotherapy in high-risk muscle-invasive bladder cancer.
